What should we look for

As a webmaster of a small site, the traffic we get (6000 unique visitors per month) is probably too small for us to be bothered; but how could we check whether the site has been compromised ?

Should we be looking for specific files, files we haven't put there, or for something else ?

Are there checks that the ISP should be carrying out ?
